It is called the LOVIT-COVID trial, and I LOVE It. Why? The trial is the first large clinical trial with 800 participants comparing placebo against intravenous ascorbic acid. The dosage will total 200 mg per kg. body weight, divided into 4 equal bolus doses in 24 hours, over a 4-day period (96 hours) for a total of 16 doses. For an adult male weighing 80 kgs, that would mean a total of 16 grams of IV C in a 24 hour period. I think they will get very decent results. I wish them the best of luck! Go Canada. Doris Loh November 27 at 4:22 PM · Why do individuals have such different responses during SARS-CoV-2? infections? One of the reasons may be due to the difference in the distribution and expression levels of the alternate receptor for the virus, CD147. Everyone knows ACE2 is the receptor that binds SARS-CoV-2. Few understand that ACE2 is found in relatively few cells and tissues, and that ACE2 is exclusively expressed at epithelial barrier sites of cells. Recent studies now question the role of ACE2 in COVID-19 respiratory tract infections because ACE2 expression levels in respiratory epithelia have been found to be extremely low [1]. In immune cells, there is no expression of ACE2 at all. On the other hand, CD147, a transmembrane glycoprotein belonging to the immunoglobulin superfamily, is widely expressed in many different types of cells, including immune cells. This receptor has many important biological functions, but it is significantly increased under pathological conditions such as cancer, as well as inflammatory diseases like COPD [8, 9], Interestingly, a study found CD147 expression to be greatly affected by age and BMI also. The authors discovered that the ability of CD147 to suppress immune responses is further enhanced in obese patients, especially those with hypertension due to increased expression of CD147 partner molecules such as cyclophilins [2]. That is probably the reason why a recent study on 1,211 European participants tested positive for C 19 and 387,079 participants who were either not tested or tested negative between March and May 2020 found that higher BMI was associated with higher odds of infection by SARS-CoV-2 [10]. In addition to immune cells, CD147 is also found abundantly in erythrocytes, commonly known as red blood cells, as well as white blood cells such as lymphocytes, macrophages [3-5]. Platelets also contain CD147 [6]. Why is this important? My peer-reviewed paper published in June discussed in depth how CD147 can cause abnormal blood clots and other vascular endothelial dysfunctions, in addition to heart damage during COVID-19 infections. The paper also showed how melatonin can attenuate all the symptoms resulting from CD147 interactions with SARS-CoV-2 [7]. Covid-19 vaccine injuries will be covered under a program known as the “countermeasures injury” compensation fund, which was set up in 2010 to cover harm resulting from vaccines for a flu pandemic, or drugs to treat an anthrax or Ebola outbreak, for example. This year, Secretary of Health and Human Services Alex Azar said the countermeasures fund should also cover injuries from Covid-19 vaccines, giving drug companies immunity from potential liability lawsuits. But the fund isn’t expected to offer much of a remedy to the public, according to lawyers and vaccine experts. SCIENCEMAG.ORG Fever, aches from Pfizer, Moderna jabs aren’t dangerous but may be intense for some Doris Loh November 24 at 1:21 PM · Today, I want to talk about potential effects of ionized lipid nanoparticles used in vaccines to transport cargo, such as viral genomic materials. Let’s start by looking at an excellent review published recently discussing the ranges of side effects that individuals experience from two mRNA vaccine candidates produced by Pfizer and Moderna. Sore arms, pains, swelling, fevers (sometimes severe), flu-like symptoms, and fatigue are among the chief complaints. However, it is believed that most people will escape “severe” side effects, defined as those that prevent daily activity, and fewer than 2% of recipients of the Pfizer and Moderna vaccines developed severe fevers of 39°C to 40°C. Interestingly, the reactogenicity,or the physical inflammatory responses to vaccination is believed to be caused by the nanoparticles in vaccines. Animals models exhibited the same type of inflammation, pain, swelling, fever, and flu-like responses when injected with nanoparticles which do not encapsulate virus mRNA genetic materials. In a study on lipid nanoparticle vaccines for the dengue virus, mice that received lipid nanoparticles alone showed statistically significant increases in IgG responses, despite the lack of viral encapsulation [2]. Both Pfizer and Moderna vaccines encapsulate CoV2 RNA genetic codes in lipid nanoparticles no larger than 0.1 micron in diameter. These oily liquid droplets serve to enclose and protect the fragile genetic instructions of the virus during manufacturing process, transportation and storage, before final injection into people [3]. Pfizer obtains its nanoparticles from Acuitas, a Canadian company, while Moderna uses a proprietary lipid technology. It is believed that not all nanoparticles are equal as the quality of the nanoparticle affects viral mRNA stability and viability. Moderna’s vaccine can survive storage for six months at -20C, the temperature of a standard domestic or medical freezer, while the Pfizer/BioNTech vaccine needs to be stored and transported at -70C. What can be the potential issues with using nanoparticles to protect viral mRNA in vaccines? Nanoparticles can cause severe oxidative stress, leading to cytotoxicity, apoptosis and DNA damage depending upon the type of lipid nanoparticle used [4, 5, 6], In general, nanoparticles (NP) enter cells via endocytosis, where they eventually end up in lysosomes that degrade them into ions. These ‘free ions’ can pass through mitochondrial membranes and react with hydrogen peroxide and oxygen to produce highly reactive and dangerous hydroxyl radicals. The generation of excess hydroxyl radicals can indirectly damage DNA, with the eventual result of cell death, or apoptosis [4]. Metallic nanoparticles, therefore, have the potential to be more toxic than lipid nanoparticles due to the magnetic charge of the free ions [10]. Even though lipids are nonpolar in that they do not exhibit either positive or negative charges like metal ions, nanoparticle technologies have developed ionizable lipid nanoparticles that are reputed to be more efficacious due to enhanced intracellular delivery [7]. The Canadian company Acuitas, has shown that the ionizable cationic lipid component in their nanoparticles critically affects the potency of lipid nanoparticle therapeutics [8]. Pfizer uses nanoparticles obtained from Acuitas [9]. It is entirely possible that side effects observed in vaccinated individuals in the Phase 3 trials for Pfizer and Moderna may have been caused in part by the nanoparticles that encapsulate viral genetic materials. One simple reason is the expression and distribution of CD147/BSG is much higher than ACE2 in many tissues and organs. Even though the expression of SARS-CoV-2 host genes are relatively stable with age, scientists recently showed that ACE2 decreases with age in some tissues but CD147/BSG actually increases with age in endothelial cells [1]. Doris Loh 7 hrs · Exactly five months ago, I published an article that explained why I believe children with symptoms of COVID toes and COVID fingers were challenged by complement activation caused by SARS-CoV-2 infections [1]. A recent peer-reviewed study found elevated levels of sC5b9 in children with SARS-CoV-2 infection, even if they have minimal symptoms of COVID-19 [2]. The authors also found that most children with SARS-CoV-2 infection had evidence of complement activation and met clinical and diagnostic criteria for thrombotic microangiopathy (TMA). Interestingly, elevated levels of sC5b9 in adults with TMA usually indicate more severe outcomes during SARS-CoV-2 infections [3]. Five months later, my views are confirmed by science. Lee et al. in a peer-reviewed study recently published in Nature Communications found no significant difference in ACE2 levels in the upper airways of patients taking blood pressure medications and those who were not [2]. Lee's study is the first mechanistic-based evidence that confirmed the use of ACE inhibitors and angiotension receptor blockers do not increase levels of ACE2 in these upper airway cells infected by SARS-CoV-2. #DorisCurve!!! So I recommended that she brush her teeth before each meal. The symptoms never returned again, unless of course, she forgot to brush her teeth. Six years later, a study showed that periodontal bacteria in dental plaques can produce enzymes and proteins that are capable of causing inflammation. These periodontal bacteria increase the risks of obesity, diabetes and metabolic syndrome. Patients with metabolic syndrome recruited in the study all tested positive for antibodies against three major strains of periodontopathic bacteria. When these bacteria were administered to mice, they all developed glucose intolerance and insulin resistance. Periodontal bacteria was also found also alter gut microbiome in the same study [1]. This is why I brush my teeth with baking soda several times daily, especially before I eat anything that involves chewing. Why am I talking about periodontopathic bacteria? Because both my favorite ancient molecules, ascorbic acid (AA) and melatonin (MEL) can attenuate periodontitis [2, 3]. There is a deep reason why melatonin is released into saliva [3], and why vitamin C intake and blood levels were found to be inversely correlated to periodontal disease in a meta-analysis involving seven cross-sectional studies that included cohort studies, case-control studies, and randomized controlled trials [2]. His best time at a meet was during his freshman year. His times had progressively become worse! Science agrees with the use of dosage escalation for patients showing severe SARS-CoV-2 symptoms requiring hospitalization. A new clinical trial with a small cohort of 66 participants sponsored by the Thomas Jefferson University will test the efficacy of IV C using Ascor® ascorbic acid in 2-hour infusions daily for 6 days. Patients with severe SARS-CoV-2 symptoms requiring hospitalization will be given escalating dosages of 300mg/kg on Day 1, 600mg/kg on Day 2, and 900 mg/kg from Day 3 to Day 6 (4 days) [2]. Melatonin supplementation is not only regarded as safe during pregnancy, science now recommends melatonin as an alternative to vaccines for the protection of mother and fetus from influenza virus infections. I have talked about the importance of melatonin for fetal development in "Light, Obesity, and COVID-19 — The Melatonin Connection" [1]. Melatonin receptors are widespread in the human embryo and fetus during pregnancy. Maternal melatonin can easily diffuse across the placenta, and is also transferred to the infant during breastfeeding. During pregnancy, melatonin levels may increase after week 24, and reach extremely high levels after week 32. Disturbances in maternal melatonin during gestation can cause metabolic alterations in the fetus [1]. Contrary to previous WHO recommendations for pregnant women as a priority group for seasonal influenza vaccination, regardless of their stage of pregnancy, evidence has now emerged showing that "anti-flu vaccines, when administered in the second or the third trimester of pregnancy, provide pregnant women with uncertain or very limited protection against influenza-like illnesses and influenza" [2] The natural increase of melatonin during pregnancy is Nature's solution to viral and respiratory infections. Doris Loh February 6 at 2:01 PM · Can melatonin cause undesired effects? This is a popular question when you scroll through comments under my melatonin posts. A simple answer is ‘No’. So where does that leave the individual who still experiences negative effects from taking pure melatonin? Melatonin is a molecule that is used by all life forms for more than 3 billion years. It has been involved with every step of evolution; which also means most biological processes would function better with the presence of melatonin. Yet for some individuals, that is clearly not the case. Let’s take rheumatoid arthritis (RA) as an example. The role of melatonin in RA has been extremely controversial, as it can actually exacerbate symptoms in RA, promoting inflammatory responses [1]. Isn't melatonin anti-inflammatory? As I have always maintained: everything in nature has the potential to be a double-edged sword; but before you make an inaccurate assumption regarding the use of melatonin in RA, let me emphasize the fact that melatonin is not Mr. Hyde here. So what is the story with MEL and RA? In the right context, even NADPH, the potent antioxidant that maintains glutathione (GSH) in its reduced form can change from Dr. Jekyll to Mr. Hyde. In rheumatoid arthritis, the context we should focus on is the famous G6PD gene that makes the glucose-6-phosphate dehydrogenase enzyme, which is the rate-limiting step in the production of the antioxidant NADPH. Individuals with glucose-6-phosphate dehydrogenase (G6PD) deficiency have a lower level of glutathione due to decreased NADPH. Is there a reason for this mutation? Why would nature want to lower NADPH/GSH? One explanation is adaptation to the malaria parasite where having lower NAPDH is protective [2]. Newer research points to something totally unexpected. Patients suffering from rheumatoid arthritis have T-cells with defective glycolytic flux because of an upregulated G6PD, Excess G6PD redirects glucose into the pentose phosphate pathway (PPP) that will increase NADPH accumulation, leading to an unbalanced redox environment with surplus reductive equivalents. This environment initiates a cascade of immune responses resulting in a hyper-inflammatory phenotype [3]. So for these individuals, increasing antioxidants may actually aggravate RA symptoms. What can be done when the G6PD gene is over- or under-expressed, leading to redox imbalance? AA is your answer. Ascorbic acid is the ultimate redox balancing molecule. An individual with deficient G6PD will benefit from AA supplementation because AA increases gene expression of G6PD. Dehydroascorbate (DHA) increased G6PD protein expression in lymphocytes by 5.9-fold, leading to a 3.3-fold increase in glutathione levels3. Treating cells with N-acetyl cysteine (NAC) only increased glutathione by 2-fold. [4]. On the other hand, excess reducing equivalents like NADPH from over-expression of G6PD will be used to regenerate oxidized AA, thus re-balancing redox environments. Studies on AA and RA dates back to the 1930’s [5], while more recent clinical data points to the efficacy of sodium ascorbate delivered in bolus IV doses in reduction of pain and fatigue in patients suffering from RA [6]. Why does SA work to reduce pain in RA? SA is a highly reactive antioxidant that can be easily oxidized and turns into a pro-oxidant that may not be easily regenerated, unlike ascorbic acid. So a bolus injection of SA will quickly oxidize GSH thus balancing the excess reductive environment created by increased endogenous antioxidants like GSH/NADPH. But what actually happens after a virus infects a host? specifically, what can a virus like SARS-CoV-2 do inside your body? In order to replicate, viruses shut down host protein synthesis in order to relocate host resources to benefit their own replication processes. This process is called 'Host Shutoff". Not only that, the virus disrupts host gene expression [1], it can also interact with host RNAs to suppress global mRNA splicing in the host, leading to to global inhibition of mRNA translation upon infection [2]. When these important cellular functions are disrupted, host immune defenses become severely compromised. Studies now show the SARS-CoV2 virus to be extremely effective and uses a broad range of tactics to inhibit host protein synthesis to suppress immune defenses[3]. The virus can degrade host mRNAs in order to take over the mRNA pool in infected cells so as to impair the translation of transcripts required for the proper expression of immune genes that normally respond to pathogen attacks [3]. This is probably one of the main reasons why some individuals have difficulty recovering from an infection, and also why I recommend continuing #lohprotocol if you still have symptoms after infection has 'cleared'. The researchers concluded that enhancement of mAb potency or adjustments to the spike sequences of current vaccines may be necessary in order to prevent loss of protection in these therapies. Because the virus can enter the nasal passages from the eyes. Is that why #lohprotocol recommends daytime MEL during infections? Yes. How does MEL protect the eyes from the virus? When airborne SARS-CoV-2 droplets land on the eye surface, the virus can enter the body through infecting the conjunctiva and travel through the nasolacrimal duct, a pathway to the pharynx [1]. A peer-reviewed study published yesterday found SARS-CoV-2 on the ocular surfaces in 52 of the 91 patients hospitalized in Lombardy, Italy as a result of CoV-2 infection. The authors detected the presence of the virus in tears collected from infected patients [2]. A peer-reviewed study from France released in late February found 7% of patients infected by SARS-CoV-2 with severe symptoms also exhibited abnormal ocular MRI manifestations, with one or several nodules of the posterior pole of the globe [3]. Another study from Hubei China found 12 of 38 patients had ocular manifestations consistent with conjunctivitis, including conjunctival hyperemia, chemosis, epiphora, or increased secretions [4]. How does SARS-CoV-2 infect the eyes? There are no studies showing the detection of ACE2 receptors are being expressed in anterior ocular tissues such as the conjunctiva or cornea. So how does the virus infect ocular surfaces if it cannot bind to ACE2 receptors? The answer may be the alternative receptor, CD147 (#lohcurve). In the eyes, CD147 (also known as EMMPRIN or Basigin) can be detected in the corneal and conjunctival epithelium, the endothelium, the stromal keratocytes, the retinal pigment epithelium, several retinal layers, and nerve fibers in the optic nerve head. What is even more interesting is that a soluble form of CD147 can be detected in the tear fluid, humor, and vitreous samples [5]. CD147 has a protective role in the eyes, regulating cell surface functions as well as peri-extracellular functions [5, 6]. Dysregulation of CD147 can lead to inflammation and dry eyes [7]. It is not surprising that the most significant ocular symptom experienced by people suffering from SARS-CoV-2 infection was sore eyes [8]. What can protect your eyes if they are infected by SARS-CoV-2? If you guessed melatonin, you get an A+. There is a reason why there are melatonin receptors in the cornea of the eyes, and why melatonin is found in tears [9-11]. Melatonin has been shown to significantly attenuate biochemical, clinical, histological, ultrastructural, and functional alterations induced by experimental uveitis which is an extremely common form of intraocular inflammation and also a major cause of visual disability [11]. The fact that melatonin is found in tears is extremely important because melatonin has been shown to bind to SARS-CoV-2 and potentially inhibit its further replicative processes [12]. If you look at the chart below, you will also see that daytime concentration of melatonin in tears is comparatively lower than that at night [10]. Since the researchers found viral presence in tears, if melatonin levels are adequate, that in itself may stop viral progression.
