Today’s exciting news about the development of an experimental vaccine called AAVCOVID, a novel gene-based vaccine candidate against SARS-CoV2, the virus that causes COVID-19, can be traced back to this couple: Jean Bennett, MD, PhD, a physician and molecular geneticist, and her husband, ophthalmologist Albert Maguire, MD.
For years, the concept of gene therapy – replacing or adding a gene to correct a disease-causing gene – was bogged down in disappointments and disasters. Nearly all failed because of the difficulties of getting a working gene into cells without also causing harmful side effects.
But Bennett and her team solved that problem by designing a virus called an adeno-associated viral (AAV) vector to deliver the gene. The virus doesn’t replicate on its own, and it doesn't cause disease in humans. Their work led to the FDA approval of the first gene therapy cure for blindness in 2017.
The promising gene-based COVID-19 vaccine in the news today uses Bennett’s team’s AAV vector to deliver genetic sequences of the SARS-CoV-2 Spike antigen so the body can develop an immune response to the coronavirus. It is currently in preclinical development with a plan to begin clinical testing in humans later this year.
Luk H. Vandenberghe, PhD, director of the Grousbeck Gene Therapy Center at Massachusetts Eye and Ear, has based the vaccine candidate on an AAV vector that he developed 15 years ago while at the University of Pennsylvania. But unlike viral vectors for gene therapy, this vector elicits an immune response, which in gene therapy is a bad thing, but in the vaccine world is ideal. It’s a brilliant innovation and a ray of hope in dark times.
